An antibody specific for PKD1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPKD1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for PKD1 is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PKD1 bound in the initial step. The color development is stopped and the intensity of the color is measured. The gene is closely linked to six pseudogenes in a known duplicated region on chromosome 16p.Polycystin-1 is a glycoprotein which contains a large N-terminal extracellµLar region, mµLtiple transmembrane domains and a cytoplasmic C-tail. It may function as an integral membrane protein involved in cell-cell/matrix interactions, and may modµLate intracellµLar calcium homoeostasis and other signal-transduction pathways. It plays a role in renal tubµLar development, and mutations in this gene have been associated with autosomal dominant polycystic kidney disease.
