An antibody specific for PXN has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyPXN present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjµgated antibody specific for PXN is added to the wells. After washing, Streptavidin conjµgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PXN bound in the initial step. The color development is stopped and the intensity of the color is measured. Turner et al. (1990) purified this protein from chicken gizzard smooth muscle, and named it paxillin ('paxillus' means 'small stake' or 'peg' in Latin) as a protein tethered to the membrane at focal adhesions.Paxillin migrates as a diffuse 65- to 70-kD band on SDS-PAGE. Salgia et al. (1995) stated that transmembrane integrin molecµLes connect the actin cytoskeleton to the extracellµLar matrix within focal adhesions.
