Test principle:This assay employs the competitive enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with an antibody specific to PF-Ab. Standards or samples are then added to the appropriate microtiter plate wells with a Horseradish Peroxidase (HRP)-conjµgated PF-Ab and incubated. The competitive inhibition reaction is launched between with HRP labeled PF-Ab and unlabeled PF-Ab with the antibody. A substrate solution is added to the wells and the color develops in opposite to the amount of PF-Ab in the sample. The color development is stopped and the intensity of the color is measured.
Product Overview:Desmoglein 1 is a calcium-binding transmembrane glycoprotein component of desmosomes in vertebrate epithelial cells. Currently, four desmoglein subfamily members have been identified and all are members of the cadherin cell adhesion molecµLe superfamily. These desmoglein gene family members are located in a cluster on chromosome 18. The protein encoded by this gene has been identified as the autoantigen of the autoimmune skin blistering disease pemphigus foliaceus.From restriction enzyme analysis, they showed that the order of the DSG genes and their orientation is as follows: 5-prime--DSG1--DSG3--DSG2--3-prime. The desmoglein isoforms are expressed in a stratification-related manner in epidermis, DSG1 being suprabasally expressed and DSG3 at a lower level, while DSG2 expression is weak and basal.
Stability:The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature.
